Description

  • About Tesamorelin (GHRH analog / Egrifta® active ingredient)

Tesamorelin — TH9507 (CAS 218949-48-5) — is a synthetic growth hormone-releasing hormone (GHRH) analog consisting of the full 44-amino acid sequence of endogenous GHRH conjugated to a trans-3-hexenoic acid group at the N-terminus, which confers enhanced stability against dipeptidyl peptidase IV (DPP-IV) degradation. As an FDA-approved GHRH analog (Egrifta®), tesamorelin stimulates pituitary somatotroph cells via the GHRH receptor (GHRHR) to increase endogenous GH pulsatility with less supra-physiological GH elevation than direct GH administration.

This makes it a pharmacologically distinct tool from direct GH and from GHRPs (ipamorelin, GHRP-2) for dissecting pituitary GHRHR signaling. Supplied as lyophilized peptide for non-clinical research use only.

    COMMON RESEARCH APPLICATIONS

    GHRH Receptor Signaling
Activates GHRHR on pituitary somatotrophs via Gs/cAMP/PKA pathway, increasing somatotroph proliferation and GH synthesis. Used in GH3 pituitary cell models to characterize GHRHR activation kinetics, receptor desensitization, and GH secretion dose-response.

    Comparative GHRH vs GHRP Signaling
Tool compound for distinguishing GHRHR-mediated GH release from GHSR-1a (ghrelin receptor) GH release. Used alongside ipamorelin and CJC-1295 to study receptor-specific contributions to pulsatile GH secretion and IGF-1 downstream signaling.

    Metabolic Body Composition Studies
Used in preclinical models examining visceral adipose tissue reduction, lipid metabolism (hepatic triglyceride, FFA flux), and insulin sensitivity as downstream effects of GH/IGF-1 axis stimulation in adipocyte and hepatocyte model systems.

    DPP-IV Stability Research
The N-terminal trans-3-hexenoic acid modification prevents DPP-IV cleavage at position 2 (Ala→modified). Used in stability studies comparing tesamorelin vs. native GHRH and CJC-1295 (DAC) for DPP-IV resistance in plasma matrix experiments.

    • What is this product?
Tesamorelin (TH9507) is supplied as a lyophilized GHRH analog peptide (CAS 218949-48-5, MW 5135.77 g/mol) for laboratory research use only. It is the active ingredient in Egrifta® (FDA-approved) and stimulates pituitary GHRHR to increase endogenous GH pulsatility.
    • What purity and testing methods are used?
Each lot is tested by reverse-phase HPLC and MS confirmation to ≥98% purity with sequence verification. A lot-specific COA is included with every order, accessible via QR code on the label.
    • What are the key chemical identifiers?
      CAS 218949-48-5 · Molecular formula C₂₂₁H₃₆₈N₆₈O₆₅S · MW 5135.77 g/mol · Synonyms: TH9507, Egrifta active ingredient, trans-3-hexenoic acid-GHRH(1-44).
    • How does tesamorelin differ from CJC-1295 and sermorelin?
Sermorelin is a truncated GHRH(1-29); tesamorelin is the full GHRH(1-44) with N-terminal modification for DPP-IV stability. CJC-1295 (DAC) additionally conjugates to albumin for extended half-life (~8 days). Tesamorelin is preferred when studying authentic GHRH(1-44) biology with improved stability but without albumin binding.
    • How is tesamorelin reconstituted for research use?
Dissolve in sterile water (pH 6–7) at ~1 mg/mL. Aliquot and store at −80°C for long-term stability. Avoid alkaline pH. Protect from light. Avoid repeated freeze-thaw cycles.
    • What is the recommended storage upon receipt?
Store immediately at −20°C desiccated. For long-term stability, aliquot and store at −80°C. Short-term at 2–8°C acceptable. Inspect vials for integrity upon receipt.
    • What is a COA and why does it matter?
A COA documents lot-specific purity (HPLC ≥98%), sequence verification (MS), and appearance. Each tesamorelin lot ships with a QR-accessible COA. Review before use to confirm identity and purity.
